    Unable to load this MKV on my UA46C5000 using Samsung C Device Profile.

    The frame rate on that file seems really strange - 719.28? Can you try recoding it using some video converter to a different frame rate and see if that helps?

    Any configurations to get Mezzmo to transcode the file?

    At the moment Mezzmo does not support transcoding based on frame rates, but it's possible to force-transcode this one file by either modifying the device profile or adjusting settings in the Advanced tab for the device - let me know which you'd prefer and I'll tell you how to change it.
